PROTECTIVE AND THERAPEUTIC ROLE OF VITAMIN B12 AGAINST SILICA NANOPARTICLES INDUCED KIDNEY AND TESTES INJURY IN RATS.

Abstract


This study was carried out to assess the effect of silica nanoparticles, Si NPs (SiO2) and vitamin B12 on the structure of renal and testicular tissues of rats. Forty adult male rats were divided into four groups. Group I served as control (Cont.), Group II served as a (protective group) (port.), was given vitamin B12 for 4 weeks and then with SiO2 for other 4 weeks. Group III was treated with SiO2 nanoparticles (SiO2) for 4 weeks and prior to that, a saline solution was given. for 4 weeks , and the last one ,Group IV was treated with SiO2 for 4 weeks and then with vitamin B12 for other 4 weeks (therapeutic group) (Thrp.). The kidney and testes specimens were prepared for staining with hematoxylin and eosin, bromophenol blue and Periodic Acid Schiff.Likewise, finally immunohistochemical detection of P53 and caspase-3 was done.
The obtained results revealed that group II (Prot.) had nearly similar findings as group I(Cont.). But SiO2- treated group showed lobulated glomerulus and pyknotic nuclei. There was interstitial hemorrhage and degenerated renal tubules. Also, degenerated seminiferous tubules with irregular shape and loss of spermatogenesis stages with few spermatocytes were observed. There was a decrease in the proteins and polysaccharide contents in both renal and testicular tissues clearly observed in the SiO2- treated group. Beside that, there were significant increases of P53 expression positive reaction in the nuclear material in renal tissue and caspase-3 positive reaction in the cytoplasm of the testicular tissue in treated group as apoptotic evidence, when compared to the control group, and Groups II

In concluded this study, the pathological examination revealed that the testicular tissue exposed to nanoparticles is more affected than renal tissue, where investigations were indicated by the immunological studies. Therefore, it is necessary for personel who have direct contact with nanoparticles to consider the safety guidelines. Vitamin B 12 must be used for their health benefits. It improves each of renal and testicular tissue functions, also keep the maintenances of carbohydrates and protein contents. It is therefore advisable to take it daily from natural sources or from tablets.
