Michaelis-constant (Km) as a factor affecting glucose up-take by some yeasts

Abstract


The present investigation aimed to study the effect of Michaelis-constant (Km) on glucose uptake by three species of yeasts fungi; Zycosaccharomyces rouxii, Zycosaccharomyces globosus and Debaryomyces hansenii.
The cells were grown either aerobically or anaerobically in the absence or presence of either glycerol or sodium chloride to regulate the water activity.
For cells grown aerobically, in case of Zycosaccharomyces rouxii, the value of Km were 62.5, 35.7 and 111.1 mM in control and in presence of glycerol or sodium chloride respectively. However, these values were 38.90 and 11.1 mM in cells grown anaerobically.
For Zycosaccharomyces globosus, the value of Km were 27.29 and 10 mM for cells grown aerobically and 64.64 and 33mM for cells grown anaerobically.
However, the values of Km were 133.3, 80 and 33.8 mM when cells of D.hansenii were grown aerobically and 74,166.6 and 58 mM for cells grown anaerobically.
Maximum velocities for glucose – uptake in presence of either glycerol or sodium chloride were higher than in their absence (control) whether the cells were grown aerobically or anaerobicall aerobically and 64.64 and 33mM for cells grown anaerobically.
